Evonetix enables Desktop Gene Synthesis

In combination with gene editing, advances in gene synthesis are critical to the success of synthetic biology companies. It increasingly finds applications in industrial biotech, materials, healthcare, and AgriTech. However, the synthesis of long DNA strands is generally slow and tune-consuming. This is why startups are working on compact and more efficient gene synthesis technologies.

British startup Evonetix develops a desktop DNA synthesis platform. The startup’s proprietary solution uses a silicon chip to control DNA synthesis. The chip utilizes micro-electromechanical systems (MEMS) technology to enable highly parallel synthesis at thousands of sites. Lastly, the strand assembly process identifies and removes any errors

SynHelix provides Large-scale DNA Manufacturing

Affordable gene synthesis facilitates building large oligonucleotide libraries to design and test synthetic gene circuits. Efforts to synthesize entire genomes from scratch are also underway. However, current gene synthesis technologies find limited successes in the manufacture of large DNA fragments. Biotech startups develop novel methods for large-scale and low-cost gene synthesis.

SynHelix is a French startup offering large-scale DNA manufacturing technology. The startup’s technology employs a specifically engineered enzyme to enable robust and scalable DNA synthesis. It allows one-step generation of long, high-fidelity DNA fragments without any limits on the maximum length. Moreover, the synthesized DNA does not require any additional downstream steps for most applications.

Camena Bioscience develops DNA Writing Tech

Current gene synthesis technologies are prone to errors. Moreover, some DNA sequences, such as those with complex secondary structures or repetitive sequences, are especially difficult to synthesize. This limits the ability of biotech startups and companies to write DNA. Biotech startups are innovating with new gene synthesis technologies for the accurate synthesis of complex genes.

US-based startup Camena Bioscience enables precise nucleic acid synthesis. The startup’s enzymatic de novo synthesis and gene assembly solution, gSynth, produces long and complex DNA molecules with high accuracy. Moreover, it allows synthetic biology companies to manufacture precise gene variant libraries for protein engineering.

OriCiro enables Cell-free DNA Synthesis

The time it takes for gene sequencing has come down by orders of magnitude in the last two decades. However, gene synthesis still takes long run times, slowing down synthetic biology workflows. This is why startups are working on cell-free DNA synthesis technologies. Moreover, these solutions use isothermal reactions to amplify DNA faster than polymerase chain reaction (PCR).

Japanese startup OriCiro provides technologies for cell-free gene synthesis and amplification. The startup’s cell-free solutions replicate the gene cloning process as it occurs in E. coli. It also develops a one-step isothermal reaction that allows the assembly of DNA fragments up to 200 kilobases long. Lastly, the amplification process provides extremely high fidelity, including for sequences with high GC content.

Helixworks manufactures Programmable DNA Sequences

As the molecule that carries evolutionary information with high fidelity, DNA offers a highly dense and durable mode of digital data storage. However, the use of DNA for data storage is presently impractical due to slow read and write times. Biotech startups are working on solutions that increase the speed of gene synthesis. This allows companies to encode digital data into DNA, for instance, to develop traceability solutions.

Helixworks is an Irish startup that builds a DNA nanotechnology platform. The startup’s platform combines programmable architectures, modular assembly, and enzymatic synthesis to write long DNA strands with minimal errors. It also develops techniques for direct sequencing of single-stranded DNA (ssDNA) and RNA, improving the accuracy of reading and writing DNA. Helixwork’s technology enables dense data storage, as well as high security and privacy.
